Process Scale Chromatography

Fundamental Question in Prep Chromatography

For successful Process Liquid Chromatography the question below have to be answered:

a. What are your sample compounds?

  1. Organic Synthetic Compounds with MW <2000 (Heterocycles, Vitamins, Organo-metallics, Hormones, Pesticides)
  2. Natural Products  with MW <2000 (Saccharides, Lipides, Phospholipides, Isoprenoids, Steroids, Alkaloids, Flavenoids, Terpenes, Polyphenols, Saponins, Proanthocyanidins, Tannins etc from a wide range of matrixes)
  3. Amino Acids, Peptides, Oligonucleotides
  4. Large scale racemate purification
  5. Inorganics Separation (Cations and Anions, Metals, Nobel Metals, Acids, Alkalis, Industrial Waste, Hydrometallurgical- refining,  leachages, extracts, precipitants)
  6. Protein, Biomarkers and Biotherapeutic Proteins (Enzymes, Membrane Proteins, Recombinant Proteins, Antibodies, extracts from mammalian Tissue, Erythrocytes, Yeast, Bacteria)

b. Sample preparation?

  1. In what condition is the compound supplied for chromatography processing?
  2. Is sample preparation necessary?
  3. What type of sample preparation is necessary?

c. Environmental factors

  1. Does the compound have to be process in organic solvent or can it be done in aqueous systems?
  2. Is ATEX configuration necessary?
  3. Are there facilities for solvent recycling?
  4. What shall happen with the by-products?

d. Process equipment selection

  1. Anticipated Business volume (g, kg or tons per day/week or month of pure compound)?
  2. Level of purity to be achieved?
  3. Envisaged process life?
  4. Envisaged process and maintenance costs
  5. Envisaged capital cost?
  6. Strategy to finance capital investment?
  7. Separation materials to be used?
  8. Has a phase evaluation been conducted to assure selection of the most effective material.

e. Preferred Process Mode

  1. .Single columns batch chromatography (Flash, MPLC, HPLC, Expanded Bed Purification, new experimental processes)?
  2. Parallel column chromatography?
  3. Continuous column processes (SMB , SSR)?
  4. New experimental processes?
