Unison Product Range

Unison series are HPLC columns for low to high polarity compound separation. The column is filled with 3 um high purity silica particles modified with octadecyl groups.

The column provides high speed analytical test results , low solvent consumption, high resolution and wide range of pH stability.

Star-up condition

Injection sample and eluent must be filtered through 0.2 um or 0.45 um porous filter to avoid column’s pressure increase.

LC Instrument set up:

To achieve maximum column performance , LC System should be set-up as follows:

  • Set peak response of detector and data processing software to maximum level for high speed analysis time (within two minutes)
  • Minimize length of tubing between injector-to-column and column-to-detector
  • When flow rate is low (e.g. 0.2ml/min), semi-micro LC system system is recommended
  • For gradient elution, a binary pump with small mixing chamber should be used

Process Conditions:

Eluent:

  • A general eluent comprises a mixture of methanol or Acetonitrile
  • Ionic solute analysis: A suitable pH modifier should be added into eluent such as TFA, acetic acid or phosphate buffer
  • Solvent: Use only HPLC grade of solvents

 Flow Rate:

  • Flow rate settings depend on column length and ID, elution time, system pressure and column temperature.
  • Standart flow rate for 4,6 mll/nin. wtef 5 un particles. If column length is longer than 15 cm fkow rate should be set lower (e.g. 0.2 ml/min to avoid exceeding the maximum pressure
  • Recommended Flow rates:
    • 1 mm ID= 0.03 - 0.2 mlmin
    • 1.5 mm ID = 0.05 - 0.3 ml/min
    • 2 mm ID = 0.1 - 0.6 ml/min
    • 3 mm ID =  0.2 - 1.0 ml/min
    • 4.6 mm ID = 0.5 - 2.0 ml/min
    • 10 mm ID = 2 - 4 ml/min 
  • High pressure columns are avail able for higher flow rates (HT and UP versions)

Pressure Limits

  • The operating maximum pressure for this column is
  • approx. 25 MPa for 3 um particles
  • HT Col Type: approx. 50MPa for 3 um particles
  • UP Col Type approx. 100 MPa for 3 um particles
  • approx. 15 MPa for 10 um particles - 28 mm ID.Column.

Column Temp

  • Operating temp is 15 - 65 C
  • Column Temp should be set considering reproducibility of retention, separation and peak shape

pH

  • The usual pH range for this column is between 2 to 7
  • After high alkali operation change eluent to storing solvent to increase column life

Column Storing Solvent

  • Initial Solvent is inspection solvent
  • The column should be stored at room temperature filled with approx 60 % Acetonitrile /40% Water or 70 % Methanol/ 30% distilled Water and have tightly screwed end plugs.
  • Please do not allow the column to dry out under any circumstances
